Cover: MD simulations on a K2P channel reveal a novel ‘C-type’ closed conformation by pulling M4 helix to allosterically modulate fi lter gating. The ‘seesaw’ movements of M4 helix induce rearrangements of networks within the fi lter region. The novel gating mechanisms help to elucidate allosteric mechanisms of ligand activation and gain‐of‐function mutations, which further support the understanding of closed state.
