Cover: To derive haploid embryonic cell lines of specific zebrafish strain, oocytes are fertilized by ultraviolet (UV)-irradiated sperm to generate parthenogenetic haploid embryos, which are dissociated at the blastula stage and cultured in vitro using the haploid culture medium containing BMP4. Gene editing in the established zebrafish haploid embryonic cell lines can be conducted through the ribonucleoprotein (RNP) transfection system. See mjad077 by Liu et al. for details.
